چکیده
Over the last 3 decades, electrochemistry (EC) has been successfully applied in phase I and II metabolism simulation studies. The electrochemically generated metabolite-like oxidation products can react with selected reagents to form conjugates. During conjugate formation, generation of isomeric compounds is possible. Such conjugates are often separated by high-performance liquid chromatography (HPLC). Here, we demonstrate a powerful approach that combines EC ion mobility spectrometry separate possible In detail, present hyphenation microfluidic electrochemical chip an integrated mixer coupled online trapped (TIMS) time-of-flight high-resolution mass (ToF-HRMS), briefly chipEC-TIMS-ToF-HRMS. This novel method achieves results several minutes, which much faster than traditional separation approaches like HPLC, was drug paracetamol controversial feed preservative ethoxyquin. analytes were oxidized situ under formation reactive intermediates mixed different thiol-containing These analyzed TIMS-ToF-HRMS identify isomers. It observed both ethoxyquin two conjugates, characterized mobilities, each reagent. Therefore, using this hyphenated technique, it not only their but also detect these within few minutes.

High resolution ion mobility-mass spectrometry for separation and identification of isomeric lipids.
Lipidomics is a particularly difficult analytical challenge due to the number and importance of isomeric species that are known or postulated in biological samples. Current separation and identification techniques are too often insufficiently powerful, slow or ambiguous. High resolution, low field ion mobility coupled to mass spectrometry is shown here to have sufficient performance to represen...
متن کاملSeparation of isomeric peptides using electrospray ionization/high-resolution ion mobility spectrometry.
In this paper, the first examples of baseline separation of isomeric macromolecules by electrospray ionization/ion mobility spectrometry (ESI/IMS) at atmospheric pressure are presented. The behavior of a number of different isomeric peptides in the IMS was investigated using nitrogen as a drift gas. The IMS was coupled to a quadrupole mass spectrometer, which was used for identification and sel...
متن کاملEvaluation of opiate separation by high-resolution electrospray ionization-ion mobility spectrometry/mass spectrometry.
The separation of opiates and the primary metabolites was evaluated with ESI-IMS/MS. Seven opiate molecules were analyzed, and spectra were shown for each compound. The IMS separation of two isomers (morphine and norcodeine) was shown with baseline separation. Differences in the mobilities were found for the nonacetylated, monoacetylated, and biacetylated compounds. In this study, two primary f...

The integration of a trapped ion mobility spectrometer (TIMS) with a mass spectrometer (MS) for complementary fast, gas-phase mobility separation prior to mass analysis (TIMS-MS) is described. The ion transmission and mobility separation are discussed as a function of the ion source condition, bath gas velocity, analysis scan speed, RF ion confinement, and downstream ion optical conditions. TIM...
متن کاملElectrospray ionization high-resolution ion mobility spectrometry-mass spectrometry.
A hybrid atmospheric pressure ion mobility spectrometer is described which exhibits resolving power approaching the diffusion limit for singly and multiply charged ions (over 200 for the most favorable case). Using an electrospray ionization source and a downstream quadrupole mass spectrometer with electron multiplier as detector, this ESI-IMS-MS instrument demonstrates the potential of IMS for...
